Drug intelligence / Profile preview
Yizekang + ivonescimab is an investigational, chemotherapy-free combination regimen consisting of two first-in-class bispecific biologics: izalontamab brengitecan (Yizekang, BL-B01D1), an EGFRxHER3-targeting bispecific antibody-drug conjugate (ADC), and ivonescimab (YiDaFang, AK112), a PD-1xVEGF-targeting bispecific antibody. Izalontamab brengitecan consists of a humanized bispecific antibody targeting EGFR and HER3 conjugated to a novel topoisomerase I inhibitor payload (Ed-04) via a cleavable linker, designed to simultaneously block EGFR/HER3 oncogenic signaling and deliver potent cytotoxicity directly to tumor cells. Ivonescimab combines immune checkpoint inhibition (PD-1 blockade) with anti-angiogenesis (VEGF inhibition) to restore T-cell activity and normalize tumor vasculature. This combination is being evaluated in Phase II/III clinical trials, including the PANKU-Lung05 and PANKU-Lung06 studies, as a first-line treatment for patients with locally advanced or metastatic squamous and non-squamous non-small cell lung cancer (NSCLC).
