Drug intelligence / Profile preview
**Izalontamab brengitecan + SI-B003** is a combination of two investigational antibody-based therapies used in oncology clinical trials. Izalontamab brengitecan is a first-in-class bispecific antibody-drug conjugate (ADC) that targets both *epidermal growth factor receptor (EGFR)* and *human epidermal growth factor receptor 3 (HER3)* and delivers a cleavable tetrapeptide-linked payload of a novel topoisomerase I inhibitor directly into targeted cancer cells, inducing DNA damage and cell death[1][2][4][5]. SI-B003, also referred to as danvilostomig in some sources, is a PD-1 monoclonal antibody immune checkpoint inhibitor. The combination is currently under investigation in multiple phase II trials for several solid tumors, including cervical cancer, gynecological malignancies, and gastrointestinal cancers, aiming to enhance antitumor efficacy by combining targeted cytotoxicity (via ADC) and immune checkpoint blockade (via PD-1 inhibition)[4][6][7].
