J65 vaccine + E39 vaccine is a combination of two synthetic peptide vaccines derived from the folate-binding protein (FBP/folate receptor alpha), which is overexpressed in several cancers including breast and ovarian cancer. - **E39** is an HLA-A2-restricted immunogenic peptide designed to stimulate cytotoxic T lymphocyte responses against tumor cells expressing FBP[1][2][7]. - **J65** (also known as E39' or GALE 302) is an attenuated version of the original E39 peptide. It was developed to reduce T cell exhaustion and improve long-term immune memory by providing a less intense but more sustainable stimulation of T cells[1][4][5]. The vaccines are typically administered with granulocyte-macrophage colony-stimulating factor (GM-CSF) as an adjuvant. The goal is to prevent recurrence in patients rendered disease-free after standard therapy for high-risk breast or ovarian cancer[1][3]. Clinical trials have evaluated both peptides alone and in sequence/combination as primary vaccination and booster regimens[8]. Both induce antigen-specific CD8+ T cell responses targeting tumor-associated antigens.
