JAB-3068 + toripalimab is a combination therapy comprising JAB-3068, an orally administered small molecule inhibitor targeting Src homology phosphotyrosyl phosphatase 2 (SHP2), and toripalimab, a monoclonal antibody immune checkpoint inhibitor targeting programmed cell death protein 1 (PD-1). JAB-3068 inhibits SHP2, a protein phosphatase involved in the RAS/MAPK signaling pathway, frequently dysregulated in cancer. Toripalimab blocks PD-1-mediated immune evasion by tumor cells, enhancing antitumor immune responses. The combination is being investigated for safety and preliminary efficacy in patients with advanced solid tumors, including non-small cell lung cancer (NSCLC), esophageal squamous cell carcinoma (ESCC), and head and neck squamous cell carcinoma (HNSCC), leveraging potential synergy between direct tumor pathway inhibition and immune checkpoint blockade[2][4][7][8].
