JAB-3312 + glecirasib is a combination therapy targeting the RTK-RAS-MAPK signaling pathway in cancer. JAB-3312 (sitneprotafib) is a potent allosteric SHP2 inhibitor with a binding Kd of 0.37 nM, while glecirasib (JAB-21822) is a selective covalent inhibitor targeting GDP-bound KRAS G12C mutations. The combination demonstrates synergistic anti-tumor effects by simultaneously inhibiting SHP2-mediated pathway activation and mutant KRAS G12C signaling. Clinical trials have shown promising efficacy in KRAS G12C-mutated non-small cell lung cancer, with objective response rates of up to 86.7% in first-line treatment settings.
