JBZ-001 + αCD38 mAbs is a combination therapeutic regimen under development by Jabez Biosciences for the treatment of relapsed or refractory multiple myeloma. The combination pairs JBZ-001, a small molecule inhibitor of dihydroorotate dehydrogenase (DHODH), with monoclonal antibodies (mAbs) targeting CD38. DHODH is a rate-limiting enzyme in the de novo pyrimidine synthesis pathway, which is often upregulated in cancer cells to support rapid proliferation; its inhibition leads to metabolic stress and cell cycle arrest. CD38 is a glycoprotein highly expressed on the surface of plasma cells in multiple myeloma, and anti-CD38 antibodies work through multiple mechanisms including antibody-dependent cellular cytotoxicity (ADCC) and direct apoptosis. This combination strategy aims to enhance the anti-tumor activity of CD38-targeted therapy by metabolically sensitizing myeloma cells through DHODH inhibition.
