JIN-A02 + amivantamab is an investigational combination regimen being evaluated in a Phase 1 clinical trial for patients with advanced EGFR-mutant non-small cell lung cancer (NSCLC) who have progressed on a third-generation EGFR tyrosine kinase inhibitor (TKI). JIN-A02 is an orally administered, fourth-generation EGFR-TKI developed by J Ints Bio that selectively and reversibly targets EGFR mutations, including the resistance mutations C797S and T790M, while sparing wild-type EGFR. It has demonstrated potent antiproliferative activity in preclinical NSCLC models and blood-brain barrier penetration. Amivantamab (brand name: Rybrevant, developed by Janssen Biotech) is an intravenous bispecific EGFR/MET monoclonal antibody that blocks both EGF and cMET receptors through ligand blocking and receptor degradation. Together, the combination dual-targets the EGFR and MET signaling pathways to overcome resistance mechanisms—particularly C797S-mediated resistance—in EGFR-mutant NSCLC.
