JK08 + pembrolizumab is an experimental combination therapy comprised of JK08—a recombinant fusion protein that contains a CTLA-4-specific antibody and an IL-15 fusion domain—and pembrolizumab, a humanized monoclonal anti-PD-1 antibody. JK08 is designed to deplete regulatory T cells and reverse immunosuppression in the tumor microenvironment via CTLA-4 targeting and stimulate natural killer (NK) cell expansion and ADCC (antibody-dependent cellular cytotoxicity) via the IL-15 domain. Pembrolizumab blocks the PD-1 receptor on T-cells, enabling antitumor immune responses. This combination is being clinically investigated in patients with advanced, unresectable, or metastatic cancers, aiming to leverage potentially synergistic immunomodulatory effects for improved clinical outcomes[1][2].
