JMT101 + FOLFIRI is an experimental combination of a recombinant humanized monoclonal antibody (JMT101) targeting the epidermal growth factor receptor (EGFR), together with the standard chemotherapy regimen FOLFIRI, which includes folinic acid (leucovorin), fluorouracil (5-FU), and irinotecan. JMT101 is being developed for the treatment of metastatic colorectal cancer (mCRC) and is used in this context to potentially enhance the antitumor efficacy of standard chemotherapy, particularly in patients with disease that has progressed after prior lines of therapy. FOLFIRI works primarily by interfering with DNA synthesis and causing DNA damage in rapidly dividing cancer cells via inhibition of thymidylate synthase (5-FU), inhibition of topoisomerase I (irinotecan), along with leucovorin to enhance the efficacy of 5-FU[1][3][5][7]. JMT101 blocks EGFR-mediated downstream signaling pathways important for tumor cell proliferation and survival.
