JMT101 + mitoxantrone liposome is an investigational combination therapy consisting of JMT101, a fully human monoclonal IgG1 antibody targeting epidermal growth factor receptor (EGFR), and mitoxantrone liposome, a liposomal formulation of the synthetic anthracycline antineoplastic agent mitoxantrone. - **JMT101:** Mechanistically, JMT101 binds to the extracellular domain of EGFR, blocking downstream signaling and recruiting immune effector mechanisms, leading to inhibition of tumor growth. JMT101 is developed as a next-generation, fully humanized cetuximab analog with higher affinity and reduced immunogenicity for EGFR targeting. - **Mitoxantrone liposome:** This is a formulation of mitoxantrone encapsulated in liposomes to enhance tumor delivery and reduce systemic toxicity. Mitoxantrone acts as a DNA intercalator and type II topoisomerase inhibitor, resulting in inhibition of DNA replication and RNA synthesis in cancer cells. The liposomal formulation appears to improve pharmacokinetics, favor tumor accumulation, and reduce cardiotoxicity compared to free drug. This combination is studied for use in solid tumors, notably including advanced colorectal cancer and breast cancer, aiming to exploit synergistic antitumor effects of targeted immune and chemotherapy.
