Overview

JNJ-42491293 + JNJ-40411813 is a combination of two investigational small molecule drugs, both developed as positive allosteric modulators (PAMs) of the metabotropic glutamate receptor 2 (mGlu2). JNJ-42491293 has been primarily used as a radiolabeled PET ligand to study mGlu2 receptor distribution and function in the brain, showing high selectivity for mGlu2 but also some binding to an unidentified site in vivo[2]. JNJ-40411813 (also known as ADX71149) is a selective mGlu2 PAM that has undergone clinical trials for neurological and psychiatric disorders including epilepsy, schizophrenia, major depressive disorder, confusion, and perceptual disorders. It was investigated for its potential antipsychotic effects and adjunctive treatment in major depressive disorder. However, development for epilepsy was discontinued by Johnson & Johnson after failed phase 2 results[6][8]. Both compounds were developed through collaborations involving Addex Therapeutics and Janssen Pharmaceuticals (a subsidiary of Johnson & Johnson)[6][8].
