This is a **combination therapy** of **JNJ-63723283** (also known as cetrelimab), a fully human IgG4 monoclonal antibody targeting programmed cell death protein-1 (**PD-1**), and **daratumumab** (marketed as **Darzalex**), a human IgG1k monoclonal antibody targeting **CD38**. JNJ-63723283 blocks PD-1, an immune checkpoint, enhancing T cell activity and anti-tumor immune responses. Daratumumab binds CD38, a cell-surface protein highly expressed on multiple myeloma cells, leading to direct tumor cell death and immunomodulation through depletion of immunosuppressive cells in the tumor microenvironment. The rationale for the combination is enhancing anti-tumor immunity by simultaneous immune checkpoint blockade (PD-1 inhibition) and direct/indirect tumor targeting (anti-CD38). The combination was under clinical evaluation in relapsed or refractory multiple myeloma but clinical development in this indication was discontinued due to lack of additional benefit and potential safety concerns in combination[1][3][5][7].
