JNJ-73763989 + JNJ-64300535 + nucleos(t)ide analogs + JNJ-64301736 is a multi-modal combination therapy regimen developed by Janssen for the treatment of chronic hepatitis B virus (HBV) infection. The regimen is designed to achieve a functional cure by targeting multiple stages of the HBV lifecycle and stimulating the host immune response. It consists of JNJ-73763989, an RNA interference (RNAi) therapeutic that silences all HBV RNA transcripts; JNJ-64300535, a capsid assembly modulator (CAM) that interferes with viral nucleocapsid formation; standard-of-care nucleos(t)ide analogs (NAs) that inhibit viral reverse transcription; and JNJ-64301736, a therapeutic DNA vaccine delivered via Ichor Medical Systems' TriGrid electroporation technology to induce T-cell responses against HBV antigens. In 2023, Janssen announced the discontinuation of its hepatitis B research and development portfolio, including this combination program.
