This is a combination therapy for chronic hepatitis B, consisting of JNJ-73763989 (also known as JNJ-3989 or ARO-HBV, a liver-targeted small interfering RNA therapeutic that silences HBV transcripts), a nucleos(t)ide analog (oral antiviral that suppresses HBV replication), and pegylated interferon alfa-2a (an immunomodulatory agent that stimulates the host immune response to HBV)[1][3][4][5][9]. JNJ-73763989 contains two N-acetylgalactosamine-conjugated siRNAs targeting HBV RNAs, leading to reduction of all HBV proteins in hepatocytes. Nucleos(t)ide analogs inhibit HBV DNA polymerase and thus suppress viral replication. Pegylated interferon alfa-2a is an engineered form of interferon that activates antiviral and immune pathways via the JAK-STAT signaling cascade. The combination is designed to induce functional cure or deep viral suppression in chronic hepatitis B patients[3][5].
