Overview

This is a **combination therapy** comprised of three elements: - **JNJ-73763989**, a short interfering RNA (siRNA) drug designed to target all messenger RNAs of hepatitis B virus (HBV), thereby reducing the expression of all viral proteins. It is administered subcutaneously and is being developed as a potential functional cure for chronic hepatitis B. - **PD-1 inhibitor**, an immunotherapy (checkpoint inhibitor) that blocks the programmed cell death protein 1 (PD-1) pathway to enhance immune responses against HBV-infected cells. This class includes drugs such as pembrolizumab or nivolumab, both monoclonal antibodies. - **Nucleos(t)ide analog**, a class of oral small molecule antivirals (e.g., entecavir, tenofovir disoproxil, or tenofovir alafenamide) that inhibit reverse transcription in HBV and suppress viral replication. The aim of this combination is to profoundly suppress HBV (and in some studies, hepatitis D virus as well), boost immune clearance of infected cells, and potentially enable a functional cure of chronic hepatitis B.
