Overview

JS009 + Toripalimab + JS006 is a triple combination therapy being developed by Shanghai Junshi Biosciences. This combination targets multiple immune checkpoint pathways to enhance anti-tumor immune responses. The combination consists of: 1. JS009 (also known as TAB009): An anti-CD112R (PVRIG) recombinant humanized IgG4 monoclonal antibody that blocks the CD112R-CD112 pathway to promote T cell activation[1][5][7]. 2. Toripalimab: An anti-PD-1 monoclonal antibody that has already been commercialized by Junshi Biosciences[5][7]. 3. JS006 (also known as TAB006): An anti-TIGIT recombinant humanized IgG4κ monoclonal antibody that blocks the TIGIT-PVR pathway[6][9]. ## Mechanism of Action This triple combination works synergistically to enhance T-cell activation through multiple pathways: - JS009 blocks the CD112R-CD112 pathway, preventing immunosuppression mediated by this interaction[5][7][10]. - JS006 blocks the TIGIT-PVR pathway, which is another immunosuppressive mechanism in the PVR family[6][9]. - Toripalimab blocks the PD-1 pathway, a well-established immune checkpoint[5][7]. Pre-clinical studies have shown that combining these agents can further increase T-cell activation and potentially improve clinical treatment efficacy compared to single or dual combinations[5][7]. The combination targets complementary immune checkpoint pathways, with TIGIT and CD112R both being members of the PVR family but having different binding sites for CD112[5][7].
