Overview
JS107 + toripalimab + oxaliplatin + capecitabine is a combination therapy regimen being evaluated for the first-line treatment of CLDN18.2-positive, HER2-negative advanced gastric or gastroesophageal junction (G/GEJ) adenocarcinoma. The regimen consists of JS107, a claudin 18.2 (CLDN18.2)-targeted antibody-drug conjugate (ADC) carrying a monomethyl auristatin E (MMAE) payload; toripalimab, a monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor; and the XELOX chemotherapy backbone, which includes oxaliplatin and capecitabine. This combination aims to leverage the targeted cytotoxicity of the ADC, the immune checkpoint inhibition of toripalimab, and the systemic effects of chemotherapy to improve outcomes in patients with high CLDN18.2 expression. Clinical data has shown promising efficacy with high objective response rates in this patient population.
