Overview

JS207 + capecitabine is an investigational combination therapy comprising JS207, a recombinant humanized bispecific monoclonal antibody targeting programmed cell death protein 1 (PD-1) and vascular endothelial growth factor (VEGF), and capecitabine, an orally administered fluoropyrimidine carbamate prodrug of 5-fluorouracil. JS207 is designed to simultaneously block the PD-1/PD-L1 immunosuppressive pathway to restore T-cell mediated anti-tumor immunity and inhibit VEGF-mediated signaling to suppress tumor angiogenesis and normalize tumor vasculature. Capecitabine acts as a cytotoxic chemotherapy agent; it is enzymatically converted to 5-fluorouracil (5-FU) in the body, which then inhibits thymidylate synthase and incorporates into DNA and RNA, leading to cell cycle arrest and apoptosis. This combination is being evaluated in Phase Ib/II clinical trials for the treatment of recurrent or metastatic nasopharyngeal carcinoma (NPC), particularly in patients who have failed prior platinum-based chemotherapy.
