JSI-1187 + dabrafenib is an investigational oral combination therapy composed of JSI-1187, a selective and potent ERK1/2 (extracellular signal-regulated kinase 1 and 2) inhibitor, and dabrafenib, a BRAF inhibitor. The combination is designed to target cancers with confirmed BRAF V600E/K or other MAPK pathway-activating mutations. By inhibiting ERK (the most distal kinase of the RAS-RAF-MEK-ERK pathway) and BRAF, the combination aims to overcome or delay resistance observed with BRAF or BRAF/MEK targeted therapies by preventing MAPK pathway reactivation. Developed for advanced or metastatic solid tumors, including but not limited to melanoma and non-small cell lung cancer (NSCLC), this combination is currently being studied in early-phase clinical trials for patients with BRAF V600 mutations or MAPK pathway alterations[1][2][3][4][5].
