Combination regimen consisting of JSKN003 (an anti-HER2 biparatopic antibody-drug conjugate), capecitabine (an oral chemotherapeutic prodrug of fluorouracil), and enlonstobart (a fully humanized, recombinant IgG4 monoclonal antibody targeting programmed cell death protein 1, PD-1). JSKN003 binds HER2 on tumor cells to deliver a topoisomerase I inhibitor payload, exerting cytotoxic effects through a bystander mechanism and is notable for improved serum stability. Capecitabine is metabolized to fluorouracil, which inhibits DNA synthesis in dividing tumor cells. Enlonstobart blocks PD-1, releasing immune inhibition and promoting T-cell activity against cancer. This combination is being studied primarily in advanced or refractory HER2-positive or HER2-expressing solid tumors, most notably in platinum-resistant ovarian cancer, breast cancer, and PD-L1-positive cervical cancers[1][3][4][6][9][10].
