Overview

JSKN016 is a bispecific antibody-drug conjugate (ADC) that simultaneously targets Trophoblast cell-surface antigen 2 (TROP2) and Human epidermal growth factor receptor 3 (HER3). Developed by Alphamab Oncology, the drug utilizes a proprietary site-specific glycosylation platform to conjugate a topoisomerase I inhibitor payload to a bispecific antibody, resulting in a homogeneous product with a drug-to-antibody ratio (DAR) of 4. JSKN016 is designed to bind to TROP2 and/or HER3 on the surface of tumor cells, leading to internalization and the subsequent release of the cytotoxic payload, which induces DNA damage and apoptosis. Additionally, the antibody component blocks TROP2 and HER3 signaling pathways. It is currently being investigated for the treatment of advanced solid tumors, with a specific focus on breast cancer subtypes, including triple-negative breast cancer (TNBC) and hormone receptor-positive (HR+)/HER2-negative breast cancer, where it is being studied in combination with the selective estrogen receptor degrader (SERD) D-0502.
