JSKN016 + carboplatin + tislelizumab is an experimental combination regimen for cancer therapy that includes three agents: - **JSKN016**: a bispecific antibody-drug conjugate (ADC) developed by Alphamab Oncology, targeting HER3 (Human epidermal growth factor receptor 3) and TROP2 (Trophoblast cell surface antigen 2). JSKN016 binds to HER3 and/or TROP2 on tumor cells, is internalized, and releases a cytotoxic topoisomerase I inhibitor, inducing apoptosis and exerting a bystander effect on neighboring cells. - **Carboplatin**: a platinum-based small molecule chemotherapeutic agent that forms DNA adducts and induces apoptosis primarily in rapidly dividing tumor cells. - **Tislelizumab**: a monoclonal antibody checkpoint inhibitor against PD-1 (programmed death-1) receptor, designed to block immunosuppressive signaling and enhance anti-tumor immune response. This combination is being evaluated for advanced malignant solid tumors, HER2-negative breast cancer, and non-small cell lung cancer, aiming to provide synergistic antitumor activity by combining targeted cytotoxicity, immunotherapy, and conventional chemotherapy[1][2][3][4][5][6].
