Overview

JSKN016 + paclitaxel is an investigational **combination therapy** comprising JSKN016, a bispecific antibody-drug conjugate (ADC) developed by Alphamab Oncology, and paclitaxel, a well-established chemotherapeutic agent. **JSKN016** targets both **HER3 (Human epidermal growth factor receptor 3)** and **TROP2 (Trophoblast cell surface antigen 2)**, entering tumor cells via target-mediated endocytosis and releasing a cytotoxic **topoisomerase I inhibitor** (TOPIi), thereby inducing apoptosis of TROP2/HER3-positive cells. The payload also produces a "bystander effect" by penetrating antigen-negative tumor cells[1][2][4][8]. **Paclitaxel** acts by stabilizing microtubules and inhibiting cell division, leading to apoptosis. This combination is being evaluated for safety and efficacy in advanced, inoperable, or metastatic **HER2-negative breast cancer** in phase Ib/II clinical trials, aiming to broaden the impact in resistant or heterogeneous solid tumors[3][5][6][8].
