JSKN016 + toripalimab is a combination therapy consisting of two investigational anticancer agents used together in clinical research. **JSKN016** is a bispecific antibody-drug conjugate (ADC) targeting **HER3 (Human epidermal growth factor receptor 3)** and **TROP2 (Trophoblast cell surface antigen 2)**, developed using a proprietary Glycan-specific conjugation platform. Upon binding to TROP2 and/or HER3 on tumor cells, JSKN016 is internalized, releasing a cytotoxic topoisomerase I inhibitor (TOPIi), inducing apoptosis in TROP2/HER3 positive tumor cells and exerting a bystander effect on nearby antigen-negative tumor cells. **Toripalimab** is a humanized IgG4κ monoclonal antibody targeting **PD-1 (programmed cell death protein 1)**, acting as an immune checkpoint inhibitor to enhance antitumor immune responses. The combination is under investigation for advanced malignant solid tumors and other cancers, aiming to harness both targeted cytotoxicity (via JSKN016) and immune-mediated antitumor activity (via toripalimab)[1][3][5].
