JSKN016 + toripalimab + carboplatin

JSKN016 + toripalimab + carboplatin is a combination therapeutic regimen being investigated as a neoadjuvant treatment for patients with resectable stage II-III non-small cell lung cancer (NSCLC). The regimen integrates three distinct therapeutic modalities: JSKN016, a bispecific antibody-drug conjugate (ADC) targeting both Human Epidermal Growth Factor Receptor 2 (HER2) and Human Epidermal Growth Factor Receptor 3 (HER3) with a topoisomerase I inhibitor payload; toripalimab, a monoclonal antibody that acts as a checkpoint inhibitor by targeting the programmed cell death protein 1 (PD-1) receptor; and carboplatin, a platinum-based chemotherapeutic agent. This combination aims to leverage the targeted cytotoxicity of the ADC, the immune-restoring properties of the PD-1 inhibitor, and the DNA-damaging effects of chemotherapy to maximize pathological response rates prior to surgical resection. The regimen is currently being evaluated in Phase 2 clinical trials sponsored by Shanghai Pulmonary Hospital.
