**JZP898 + pembrolizumab** is a combination therapy under investigation for advanced or metastatic solid tumors. JZP898 is a conditionally activated prodrug of interferon alpha (IFNα) engineered to preferentially deliver IFNα to the tumor microenvironment, aiming to maximize antitumor effects while minimizing systemic toxicity commonly associated with IFNα therapy. Pembrolizumab is an anti–programmed cell death protein 1 (PD-1) monoclonal antibody immune checkpoint inhibitor that promotes immune-mediated tumor cell destruction. The combination is being evaluated to exploit synergistic immunostimulatory effects: JZP898 induces direct tumor cell apoptosis and stimulates immune activation via interferon signaling, while pembrolizumab blocks PD-1, overcoming tumor-induced immune suppression. The approach seeks to provide durable antitumor activity with reduced toxicity, and is currently being evaluated in a phase 1, first-in-human, open-label multicenter trial (NCT06108050) in adult patients with advanced or metastatic solid tumors[1][3][5][7].
