Overview

KAZ954 + NIR178 is an experimental combination of two investigational cancer immunotherapies evaluated for advanced or metastatic solid tumors. - **KAZ954** is a monoclonal antibody targeting the ectonucleoside triphosphate diphosphohydrolase-2 (ENTPD2, also known as CD39L1), an enzyme involved in generating immunosuppressive adenosine in the tumor microenvironment. By blocking ENTPD2, KAZ954 is designed to activate the immune system within tumors and promote anti-tumor immunity[1]. - **NIR178** (also called taminadenant or PBF509) is an oral small molecule antagonist of the adenosine A2A receptor, expressed on T cells and other immune cells, where adenosine engagement normally suppresses anti-tumor responses. Blocking A2A receptor activity is intended to enhance T cell activity against cancer cells[1][6]. The combination seeks to reduce immunosuppression in the tumor environment through two mechanisms: (a) limiting adenosine production (via ENTPD2 inhibition) and (b) blocking adenosine’s immunosuppressive action (via A2A receptor blockade). This drug combination was tested in an open-label, multi-center, early phase (Phase I/Ib) trial sponsored by Novartis to determine safety, dosage, and preliminary efficacy in adults with advanced solid tumors that are not amenable to curative surgery[1][2][3][4][5]. Development for this combination stopped early due to business reasons, not safety concerns. There are currently no plans for further trials as of the last available reports[1].
