KB707 + Opdualag is an investigational combination therapy comprising KB707, a genetically modified herpes simplex virus type 1 (HSV-1) designed to deliver interleukin-12 (IL-12) and interleukin-2 (IL-2) directly to the tumor microenvironment, and Opdualag, a fixed-dose combination of the immune checkpoint inhibitors nivolumab (anti-PD-1) and relatlimab (anti-LAG-3). Developed by Krystal Biotech in collaboration with Bristol Myers Squibb, the therapy was evaluated via intratumoral injection for the treatment of unresectable or metastatic melanoma and other advanced solid tumors. The mechanism involves viral-mediated cytokine expression to stimulate local anti-tumor immunity alongside systemic checkpoint blockade to overcome T-cell exhaustion. In August 2025, Krystal Biotech discontinued the development of the injected formulation for solid tumors and melanoma following regulatory shifts in the oncolytic virus landscape.
