A combination therapy consisting of ketamine and rapamycin. Ketamine is an NMDA receptor antagonist with rapid antidepressant effects, while rapamycin is an mTORC1 inhibitor. When administered together, rapamycin appears to prolong the antidepressant effects of ketamine. In clinical trials, pretreatment with rapamycin (6 mg) before ketamine infusion (0.5 mg/kg) resulted in higher response and remission rates at two weeks compared to ketamine alone.
