KGX105 and KGX106 are conditionally active, protease-sensitive bispecific T-cell engagers (TCEs) developed by Kangabio for the treatment of solid tumors. KGX105 is an EGFRxCD3 bispecific antibody designed as a prodrug; its binding domains for both EGFR and CD3 are masked to prevent systemic T-cell activation and off-tumor toxicity in normal tissues. Upon entering the tumor microenvironment, tumor-specific proteases cleave the masks, allowing the antibody to simultaneously bind EGFR on tumor cells and CD3 on T cells, thereby inducing localized T-cell mediated cytotoxicity. KGX106 follows a similar design targeting other tumor-associated antigens (TAAs). Both molecules incorporate an albumin-binding domain to enhance their pharmacokinetic profile and are currently in preclinical development.
