KHK2455 + avelumab is an investigational **combination therapy** for cancer that combines two agents: **KHK2455**, a selective small molecule inhibitor of indoleamine 2,3-dioxygenase 1 (**IDO1**), developed by Kyowa Kirin, which inhibits tryptophan metabolism in the tumor microenvironment to suppress tumor immune evasion; and **avelumab**, a human monoclonal antibody targeting programmed death ligand 1 (**PD-L1**), developed by Merck KGaA (Darmstadt, Germany) and Pfizer, that blocks the PD-L1/PD-1 axis to reactivate T cell-mediated antitumor immunity and can trigger antibody-dependent cellular cytotoxicity (**ADCC**)[2][6]. This combination aims to enhance immune-mediated anti-tumor responses in subjects with advanced solid tumors, particularly advanced or metastatic urothelial carcinoma, and is currently being evaluated in early phase clinical trials[1][6][7].
