This experimental autologous dendritic cell vaccine was developed by Dr. Nina Bhardwaj for the treatment of high-risk resected melanoma. The therapeutic approach involves isolating a patient's own dendritic cells (DCs) and maturing them ex vivo while pulsing them with five specific melanoma-associated peptides and Keyhole Limpet Hemocyanin (KLH). KLH serves as a potent immunogenic carrier and adjuvant to enhance the overall immune response and acts as a tracer to monitor immune competence. The matured, antigen-loaded dendritic cells are administered intradermally to the patient, where they migrate to the lymph nodes to prime and activate cytotoxic T lymphocytes (CTLs) against melanoma cells expressing the targeted antigens. This vaccine was evaluated in Phase 1 clinical trials to compare the efficacy of dendritic cells versus standard adjuvants like Montanide in inducing anti-tumor immunity.
