KLH + peptides + Montanide is a combination immunotherapeutic formulation used primarily as a vaccine platform for inducing robust immune responses against specific peptide antigens. In this approach, synthetic peptides (often tumor- or disease-specific) are chemically conjugated to keyhole limpet hemocyanin (KLH), a highly immunogenic carrier protein. The resulting KLH-peptide conjugate is then emulsified with Montanide ISA 51 VG, an oil-based adjuvant that enhances the immune response by promoting antigen presentation and sustained release at the injection site. This combination is designed to elicit strong humoral and cellular immunity—particularly high-titer antibody production and T cell responses—against otherwise weakly immunogenic peptides. It has been investigated in preclinical models and clinical trials for cancer (e.g., melanoma, leukemia), autoimmune targets, and infectious diseases[1][2][3][4][5]. The mechanism of action involves enhanced antigen uptake by dendritic cells due to the adjuvant effect of both KLH (as carrier) and Montanide (as depot-forming adjuvant), leading to activation of CD4+ helper T cells as well as cytotoxic CD8+ T cells specific for the target peptide[1][2][7].
