KN026 + KN046 is a combination of two bispecific monoclonal antibodies developed by Alphamab Oncology (Jiangsu Alphamab). KN026 is an anti-HER2 bispecific antibody that binds two non-overlapping epitopes on the HER2 receptor, resulting in dual blockade of HER2 signaling. It demonstrates potent antibody-dependent cell-mediated cytotoxicity and antiproliferative effects against HER2-overexpressing tumor cells, including those resistant to trastuzumab or with low/medium HER2 expression. KN046 is a humanized IgG1 Fc-fused bispecific antibody targeting both PD-L1 and CTLA-4, designed to block immune checkpoints and deplete regulatory T cells within the tumor microenvironment. This dual mechanism enhances antitumor immunity by increasing effector T-cell activity while reducing immunosuppressive components. The combination has shown promising efficacy and safety in clinical trials for various solid tumors, particularly in HER2-positive cancers such as gastric cancer, colorectal cancer, and breast cancer[1][2][3][4][6].
