KN035-ELP(V) is a genetically engineered fusion protein combining the KN035 nanobody, which specifically binds to human programmed cell death protein ligand 1 (PD-L1), with a thermosensitive elastin-like polypeptide (ELP(V)). This design creates a super-long-acting anti-PD-L1 nanobody that forms a sustained-release depot after intraperitoneal injection, extending its circulating half-life and enhancing tumor accumulation and penetration. It is under investigation primarily for cancer immunotherapy, notably triple-negative breast cancer (TNBC), and shows improved antitumor efficacy and reduced adverse effects compared to KN035 and KN035-Fc. Its mechanism centers on efficient immune checkpoint blockade by inhibiting the PD-1/PD-L1 interaction, leading to durable antitumor immunity[1].
