**KN5601** is an investigational, off-the-shelf dual-target chimeric antigen receptor natural killer-cell therapy developed by Rui Therapeutics. The engineered NK cells recognize **CD19** and **B-cell maturation antigen**, enabling depletion of pathogenic CD19-positive B-cell populations and BCMA-positive plasmablasts or plasma cells. It is being evaluated after lymphodepleting chemotherapy in early-phase studies of B-cell-mediated autoimmune diseases, including IgA nephropathy, systemic lupus erythematosus, nephrotic syndrome, IgG4-related disease, and refractory pediatric rheumatic diseases.
