KRAS (G12D) A*11 T cell engager is a bispecific antibody in discovery-stage development by Myrio Therapeutics for the treatment of KRAS G12D-mutant solid tumors, including lung, pancreatic, and colon cancers. The therapeutic is designed to recognize the KRAS G12D neoantigen peptide specifically when presented by the HLA-A*11 MHC class I molecule. By simultaneously binding this tumor-specific complex and the CD3 receptor on T cells, the engager facilitates the formation of an immunological synapse, leading to T-cell activation and the targeted destruction of cancer cells. This approach aims to overcome the challenges of targeting intracellular mutations like KRAS by utilizing the cell's natural antigen presentation machinery.
