Overview

KRAS G12V TCR-T cells (Clone 5) is an adoptive cell therapy consisting of T cells engineered to express a T-cell receptor (TCR) specific for the KRAS G12V mutation presented by HLA-A*11:01. Developed by Shanghai Xinpu BioTechnology in collaboration with Ruijin Hospital, the TCR was isolated from a patient with advanced non-small cell lung cancer (NSCLC) who responded to a KRAS G12V mRNA vaccine. Clone 5 is one of several identified clonotypes that demonstrate CD8-dependent binding and high functional avidity. In preclinical studies, Clone 5 TCR-T cells showed potent cytotoxicity against KRAS G12V-mutant pancreatic and lung cancer cells in vitro and mediated significant tumor regression in xenograft mouse models. While Clone 1 was selected as the lead candidate for clinical translation due to its safety profile, Clone 5 remains a characterized candidate with a distinct peptide recognition motif.
