KRAS mutant antigen specific TCR-T cells are an autologous T-cell receptor engineered T-cell (TCR-T) therapy developed by Sun Yat-Sen Memorial Hospital of Sun Yat-Sen University. This cell therapy is designed to target specific mutations in the KRAS protein, specifically the G12V and G12D variants, which are common oncogenic drivers in various advanced solid tumors. The treatment process involves isolating a patient's own T cells and engineering them to express a specific TCR that recognizes KRAS mutant peptides presented by HLA molecules on the surface of cancer cells. Following a lymphodepletion conditioning regimen of cyclophosphamide and fludarabine, the engineered cells are reinfused into the patient, often accompanied by interleukin-2 (IL-2) support to promote cell survival and expansion. The therapy aims to induce a targeted immune response to eradicate tumor cells harboring these specific KRAS mutations while sparing healthy tissue.
