This is a personalized neoantigen-based mRNA vaccine developed by Sichuan University for the treatment of advanced solid tumors harboring KRAS mutations. The vaccine platform is designed to deliver mRNA encoding specific KRAS-mutant neoantigens to stimulate the patient's immune system, specifically activating KRAS-specific CD4+ and CD8+ T-cell responses. The therapeutic goal is to convert 'immune-cold' tumors into responsive ones, potentially enhancing the efficacy of concurrent immunotherapies. It is currently being evaluated in a Phase 1 clinical trial (NCT06344156) as a monotherapy for various KRAS-mutant malignancies and as a neoadjuvant therapy in combination with the anti-PD-1 checkpoint inhibitor toripalimab and chemotherapy (pemetrexed and carboplatin) for lung adenocarcinoma. The trial, conducted in China, began in April 2024 and is expected to conclude in 2027.
