KTN3379 + erlotinib is a combination therapy under investigation mainly for cancer treatment, particularly for solid tumors including non-small cell lung cancer and head and neck cancer. **KTN3379** is a human monoclonal antibody that blocks ErbB3 (HER3), a receptor tyrosine kinase involved in tumor survival pathways and resistance to targeted therapies. **Erlotinib** is a small molecule, reversible tyrosine kinase inhibitor (TKI) targeting the epidermal growth factor receptor (EGFR). The rationale for the combination is to inhibit redundant signaling through ErbB family members (ErbB3 and EGFR), which may enhance antitumor activity and delay resistance associated with EGFR inhibition alone. KTN3379 has been studied in combination with several EGFR pathway inhibitors, including erlotinib, to treat cancers expressing ErbB3 and/or sensitive to EGFR blockade. The combination is being developed by Kolltan Pharmaceuticals in early clinical trials, especially Phase 1b studies, focusing on dose escalation and assessment of efficacy and tolerability in patients with advanced solid tumors[1][2].
