KTN3379 + trastuzumab is a combination of two monoclonal antibodies targeting receptor tyrosine kinases implicated in cancer cell growth and treatment resistance. **KTN3379** is a fully human anti-ErbB3 monoclonal antibody with engineered YTE half-life extension, locking ErbB3 in an inactive configuration to inhibit tumor growth and overcome resistance mechanisms. **Trastuzumab** is a recombinant humanized IgG1 monoclonal antibody that binds to the extracellular domain of human epidermal growth factor receptor 2 (HER2/ERBB2), inhibiting downstream signaling pathways responsible for cancer cell proliferation and survival. This combination is designed to block both ErbB3 and HER2 signaling in HER-family driven cancers, notably overcoming resistance to anti-HER2 therapies. KTN3379 was developed by Kolltan Pharmaceuticals, and the combination has been studied in phase 1b clinical trials for safety, pharmacokinetics, and preliminary efficacy in various solid tumors[1][3][5].
