Overview

L-650,719 is a **carbonic anhydrase inhibitor** of the sulfonamide class, chemically named 6-hydroxybenzo[b]thiophene-2-sulfonamide. It was investigated as a topical agent to lower intraocular pressure for glaucoma therapy. L-650,719 is structurally characterized by a benzo-thiophene core with a sulfonamide group and hydroxyl substitution, conferring improved water solubility over earlier analogs. Topical administration in animal models lowered intraocular pressure, likely through carbonic anhydrase inhibition in ocular tissues. Its development was discontinued after clinical trials showed only modest efficacy in humans.[1][3][5][7]
