L-asparaginase + erwinia asparaginase is a **combination drug** consisting of two asparaginase enzymes, both used as antineoplastic agents in the treatment of acute lymphoblastic leukemia (ALL) and lymphoblastic lymphoma (LBL). L-asparaginase is typically derived from Escherichia coli, while erwinia asparaginase is produced from Erwinia chrysanthemi. Both enzymes deplete asparagine, an amino acid necessary for the survival of leukemic cells, by catalyzing its hydrolysis to aspartic acid and ammonia. Leukemic lymphoblasts are unable to synthesize asparagine sufficiently and undergo apoptosis when asparagine is depleted. Erwinia asparaginase is often used as a substitute for patients who develop hypersensitivity to E. coli-derived asparaginase. Both forms are administered by intravenous or intramuscular injection as part of multi-agent chemotherapy regimens[1][2][3][4][5][6][7].
