L-IFPTA+ is a nanoliposomal anti-PCSK9 vaccine designed to induce a long-lasting immune response against proprotein convertase subtilisin/kexin type 9 (PCSK9), leading to the generation of PCSK9-specific antibodies that inhibit PCSK9-LDL receptor interaction. This vaccine increases hepatic low-density lipoprotein receptor expression, resulting in significant reductions of total and LDL cholesterol in hypercholesterolemic mouse models. The formulation integrates AFFITOPE mimotope peptides, nanoliposomes, and an alum adjuvant. Demonstrated efficacy includes durable antibody response, atheroprotection, and increased Th2/IL-4 anti-inflammatory responses, with preclinical evidence in mice and non-human primates. Proposed for prevention and long-term management of dyslipidemia and atherosclerosis, clinical data beyond preclinical models are not yet available[1][2][3][4][6].
