**L19-IL2 + dacarbazine** is a combination therapy for metastatic melanoma comprising an immunocytokine fusion protein (L19-IL2) and the classic alkylating agent dacarbazine. L19-IL2 consists of an antibody fragment (L19) targeting the EDB domain of fibronectin—a marker of tumor angiogenesis—and human interleukin-2 (IL2). This design delivers IL2 selectively to tumors, promoting local immune stimulation. Dacarbazine is a cytotoxic small molecule with antineoplastic activity. Phase I/II and randomized phase II studies have demonstrated that this combination is tolerable and shows activity (e.g., improved overall response rate and progression-free survival) in advanced or metastatic melanoma[1][2][3][4].
