Overview

**laduviglusib + sodium valproate** is a combination of two small molecule drugs: laduviglusib, a selective glycogen synthase kinase-3 (GSK-3) inhibitor, and sodium valproate, a widely-used antiepileptic agent with mood-stabilizing properties. This specific combination has been studied as an investigational regimen, most notably in clinical trials for sensorineural hearing loss. Laduviglusib acts by inhibiting GSK-3, a serine/threonine kinase involved in numerous cellular processes, including neural and hair cell regeneration. Sodium valproate operates primarily by increasing brain gamma-aminobutyric acid (GABA) levels and modulating neuronal excitability. Together, these agents are hypothesized to have a synergistic effect in promoting recovery or protection of sensory hair cells in the inner ear, potentially restoring hearing in sensorineural hearing loss. The combination is studied primarily as an intratympanic (middle ear) injection in clinical settings[6][8].
