LAE001 + ADT refers to a combination therapy where LAE001, a small molecule oral dual inhibitor of CYP17A1 (steroid 17-alpha-hydroxylase) and CYP11B2 (aldosterone synthase), is administered alongside androgen deprivation therapy (ADT) for the treatment of metastatic castration-resistant prostate cancer (mCRPC)[1][3]. LAE001 blocks both androgen and aldosterone synthesis, distinguishing it from other androgen synthesis inhibitors by potentially allowing for administration without concomitant steroids such as prednisone. LAE001 is in-licensed by Laekna Therapeutics from Novartis and is the only dual CYP17A1/CYP11B2 inhibitor in advanced clinical trials for prostate cancer[1][3]. In clinical combination, LAE001 is typically given with ADT—most commonly the use of medical or surgical castration to suppress endogenous androgen production. The combination shows promise for improving progression-free survival with tolerable adverse effects in patients resistant to previous lines of standard-of-care therapies[2][3].
