Drug intelligence / Profile preview
"Lamivudine + adefovir vs entecavir" refers to a Phase 4 randomized clinical trial (NCT00531167) sponsored by Korea University that compared two rescue strategies for patients with lamivudine-resistant chronic hepatitis B (CHB). The study evaluated the efficacy of adding adefovir dipivoxil (10 mg/day) to ongoing lamivudine (100 mg/day) therapy versus switching to entecavir monotherapy (1.0 mg/day). All three agents—lamivudine, adefovir, and entecavir—are nucleoside/nucleotide analogue antivirals that function by inhibiting the hepatitis B virus (HBV) DNA polymerase/reverse transcriptase. The trial results indicated that the combination of lamivudine and adefovir provided superior virological response rates and lower rates of genotypic resistance compared to switching to entecavir monotherapy in this specific treatment-refractory population.
