Overview

Triomune Baby is a pediatric fixed-dose combination antiretroviral therapy developed and manufactured by Cipla Ltd. It contains three small molecule active ingredients: stavudine (6 mg), lamivudine (30 mg), and nevirapine (50 mg). Stavudine and lamivudine are nucleoside reverse transcriptase inhibitors (NRTIs) that act as chain terminators, while nevirapine is a non-nucleoside reverse transcriptase inhibitor (NNRTI) that binds allosterically to the viral reverse transcriptase. Together, they synergistically inhibit Human Immunodeficiency Virus type 1 (HIV-1) reverse transcriptase to suppress viral replication. Triomune Baby was specifically designed as scored, dispersible minitablets for HIV-infected infants and children weighing less than 10 kg, offering a simpler, more affordable, and more stable alternative to liquid formulations or split adult tablets. Although widely used in resource-limited settings like Malawi and Zambia, the regimen has largely been phased out or discontinued in favor of safer, newer antiretroviral combinations due to stavudine-associated toxicities.
